I've two blueberry bushes in pots, they need potting up into bigger pots soonish.

They both seem to have put up a great tall stem with leaves on, almost like a vegetable looks like when it bolts.

They're also green fly infested.

I'm going to repot them, soft soap them, but;

a) do they need pruning (not sure how old they are)

and

b) can I take cuttings from them for new bushes?

They should bush up naturally.
One or two of mine where like yours but thickened up subsequently.
They will propagate from hardwood cuttings but the success rate is low so don't expect every cutting to root.
Wood about pencil thick is best, 6 to 12" long.

On a similar note, I appear to have a seedling blueberry at the bottom of one of my pots. Is it likely to be any good do you think?

They will grow from seed with variable results but what you may have is a root sucker not a seedling.

Ok, thanks. It seems to have a root system completely independent of the parent plant.
